From what I understand the "stack-up" is the layers of the FR4, copper, resist etc
I placed an order with my PCB manufacturer and they sent this;
Asking, " should we ignore below stack-up of 2 layers.".
What does this mean?

I think they are asking if you are serious about those specs, or they can just do whatever they normally do for 1.6mm PCBs.
But better clarify with them.
